I have read that the maximum acceptable deflection in beams in a building is calculated as L/300, L/250, L/200, L/150, etc.

Uncertain about what the standard is today for steel and wood beams (glulam, construction timber...).
 
Isn't it the actual construction and what it is intended for that determines deflection and other flex, not the material used?

Exactly, it's not possible to say generally. Then there are more things that can happen than just a beam bending, depending on the support conditions, profile, etc.
