I'm building an isolated shed mainly for summer use that will remain empty and unheated in the winter (and most of the summer), but with the option to heat and sleep in it on some weekends regardless of the season.

Should I use a plastic/vapor barrier inside the insulation in the walls and roof, or should I skip it?

The ventilation will consist of (I assume) cut-out holes in the wall with a kind of disc vent?

The answer here is usually that since it will be mostly unheated, you should not have a vapor barrier.
 
Or turn it around: unheated furnished "friggebodar" tend to "smell musty". You might want to keep it slightly heated to 5-10°. With a little more insulation, it might be a question of 30-40Wh spread over a year. A lit light bulb, in other words.
