I have a hole down into the intermediate floor where the chimney goes up further to the attic. This will later be enclosed with closet walls, and a panel that can be lifted off for inspection.

But my question is:
Can I cover the hole in some way so I can avoid a lot of dust until the day I make the walk-in closet? Like cutting out a plasterboard and sealing it with something fireproof, or does it have to be open down there so the chimney sweep can inspect it? Chimney pipe passing through a hole in the floor, surrounded by insulation material, with wooden framing around the base.

As long as the chimney sweep hasn't checked the chimney and you don't light a fire, it doesn't matter how you do it. Just make sure you can remove it when the chimney sweep comes. I would call the chimney sweep and ask with what material it's most suitable to cover the hole, so you can keep it even when you start lighting a fire.
