On the walls, there is rough paneling everywhere except at the laundry chute. They have built the chute into the chimney, so it's a "column" with a plastered brick wall with a hole in the middle (we have removed the door).
I have constructed a unique structure of 2x2s and planned to build a wooden cover.
I'd like to plaster over this wooden cover along with the rest of the "column."
How do I get the plaster to adhere to the wood (rough paneling)?
Can I attach some form of mesh or something to the "cover" so the plaster adheres to it?

Why not seal the opening with bricks?

I would probably rather put a gypsum board and plaster on it afterwards. Use gypsum plaster to fill it up so the "depth becomes the same as the rest of the wall first. It should be sturdy enough so that it doesn't crack later when you plaster the whole wall. To be on the safe side, you can also place a plaster mesh over the hatch as anatis suggested.
