I have a warm garage + carport where I plan to build a loft that partially extends over the carport. I will therefore cover parts of the truss's lower frames with plastic and then lay the floor.

Question: Is an air gap needed between the plastic and the loft's floor, or can I lay the 34 mm tongue and groove boards directly on the plastic...?

You can lay the floor directly on the plastic.
