I am considering replacing old, tired wood columns with brick columns. How can I best handle reinforcement when I don't have the natural cavity in the middle since I'm choosing to build two bricks per layer? Reinforcing rods and drilling through the bricks? Any other general tips?

Yes, I think so. However, you still won't be able to handle any lateral load.
