I have built my own staircase and now need to attach the oak steps. The oak steps are cut from a countertop and will be attached (as a covering) against a standard 45x220 beam.

I could indeed screw in most of the steps, but it is difficult to access the lowest ones.

Would you have chosen regular wood glue or perhaps PL400? (which, according to the tube, fills any unevenness, etc...)

Screw visibly, hiding with stylish oak plugs is not an option?

Well... probably want to try screwing and gluing in the hidden...

then I would have chosen PL 400.. very good glue

PL400 is a very good adhesive that works excellently for your purpose.

Nice to hear! (bought a couple of tubes the other day...)
I'm using PL400 and some screws where it fits.
