A beam should be mounted against a vertical parallel beam. Can I let it reach all the way down to the concrete floor if I put some wind barrier material between the concrete and the beam to prevent moisture migration, or should I leave a space of a few cm between the beam and the floor?

If you connect wood directly against concrete, you should use some form of insulation ("paper") that is sufficiently vapor-tight. The wind barrier is too open to be used for this purpose.
