I will be replacing 2 swing gates with a roll-up door in my garage. The person who originally built the garage left photo documentation of how it was built. The door opening will be enlarged along the red lines, but somewhat straighter.

Do I need to reinforce the existing construction in any way beyond the new door frame?

Check with what the supplier of the port says. Which port is it about?

Referring to the durability of the building construction since studs disappear.
My own opinion is that the durability is not affected. It has nothing to do with the installation of the door.
 
I think the wall looks strange and probably doesn't support anything. There should have been a W truss at the end as well, like further in. It is self-supporting, and you can basically remove the entire wall. The roof will have less to hang on as you remove more, given how it's currently built.
 
Place a glued laminated timber beam horizontally with proper pillars. That is adapted to the span and load from the roof. Check the träguiden.
 
The outermost chair probably doesn't have a W compartment (?) so it will need support...
